Advanced prostate cancer biomarker discovery with proteomic technology
-Using 2-dimentional difference in gel electrophoresis system and mass spectrometry, analyzed proteins from grade 4/5 prostate cancer and BPHs. Identified 100 interesting proteins (some unreported) as potential markers. ELISA was performed in hundreds of clinical sera.
-Ran proteins from frozen tissues and sera. Applied laser-capture microarray technology for frozen sections, and affinity column for removal of abundant proteins in sera.
-Collaborated with ABI and SIGMA in high volume cancer serum and high volume BPH analysis with the depletion of top 20 abundant proteins and quantitative analysis with 4800 Plus MALDI TOF/TOF™ Analyzer.
-Collaborated with Ciphergen for SELDI-TOF analysis for total proteins from LCM treated cancer cells and BPH cells in 2000.
-Presented results in international meetings including AACR Annual Meeting, HUPO Annual Meeting and Mass Spectrometry Annual Meeting.

Advanced prostate cancer biomarker discovery with microarray technology
-In very successful collabration with Affymatrix, screened the global gene expressions from different categories of prostate cancers, as well as BPH, dysplasia and normal with 6500 FL chips and U133 chips.
-In combined effort of Dr Stamey, who introduced PSA for the diagnostics of prostate cancer, and Dr John McNeal, the top pathologist of prostate cancer, characterized differences in gene expression from different histological zones: peripheral zone, transitional zone, central zone and seminal vesicle.
-Awarded 2 patents.
-4 papers published.
-First to report hepsin and maspin were inversely expressed in cancers. Research highly praised by former NCI Director Dr. Andrew C. von Eschenbach. Our unique database is still used in identifying interesting genes for clinical trials.

Early diagnostic biomarker discovery of bladder cancer with DNA genetic polymorphism analysis
-In international collaboration with JHU and MD Anderson Cancer Center, performed genomic DNA polymorphism research.
-Screened hundreds of microsatellite DNAs throughout human genome by PCR in human bladder cancers, both with urine and matched tissues, to find early biomarkers for diagnostics.
-Employed isotope labeling and manual microdissection.
-Findings published in journals of J.NCI and Can Letters.
-Work awarded “one of the top ten achievements in cancer research” in 2000 by the Ministry of Human Health.

The genetic instability analysis in the process of myelodysplastic syndromes evolving into leukemia
-Member of team performing pioneering research of genomic instability study using PCR in the process of MDS (pre-leukemia) evolving into acute myeloblastic leukemia.
-Bcl-2 and c-erbB2 expression in MDS by in situ hybridization and immunocytochemistry.
-Findings published in leukemia.
