One major bottleneck in the generation of many antibody therapeutics is the secretion of biologically active immunoglobulins (Ig). It is believed that the regulation of Ig secretion is the rate-limiting step in antibody production. Signal transduction pathways tightly control the physical movement and secretion of Ig protein from the intracellular to the extracellular space. The Rho GTPase signaling pathway is a family of G proteins that have been shown to regulate many aspects of intracellular functions. Rho GTPases exert profound effects on cytoskeletal dynamics which ultimately regulate vesicular transport of secretory proteins. Ig is a major secretory protein regulated by the Rho GTPase family. By utilizing published information from two antibody secretion cellular model systems (B cells and polarized epithelial cells) this project will delineate the contributions of the Rho GTPase signaling pathway on Ig secretion in CHO and NS0 cells.
